Case Report
An otherwise healthy 23-year-old man was brought to the emergency room after the onset of two tonic–clonic seizures, witnessed by his friends. Indeed, while he was attending a pool party, he presented with a sudden loss of consciousness and generalized tonic–clonic seizure, followed after around 30 min by another similar episode.
While in the emergency room, his general and neurological examinations as well as laboratory tests and urinalysis, including alcohol, cocaine, opioid, cannabinoid, LSD (d

Mechanisms underlie the proconvulsant effects of sildenafil
2021, Biomedicine and PharmacotherapyProconvulsant effects of sildenafil citrate on pilocarpine-induced seizures: Involvement of cholinergic, nitrergic and pro-oxidant mechanisms
2019, Brain Research BulletinCitation Excerpt :In humans, sildenafil related seizures were firstly described in two adult healthy men after the use of a prescribed dose of 50 mg for erectile dysfunction (Gilad et al., 2002). Also, the occurrence of spontaneous seizures in non-epileptic patients were described after the use of other PDE inhibitors, such as tadalafil (Calabrò et al., 2013) and vardenafil (Koussa et al., 2006; Striano et al., 2006) at prescribed doses, strengthening the proconvulsant potential of this class of drugs (Matos et al., 2012). Furthermore, the high ingestion of caffeine, a methylxanthine and nonselective PDE inhibitor, is related to increased seizure frequency in epileptic patients, indicating that PDE inhibitors should be carefully used by epileptic patients (Chrościńska-Krawczyk et al., 2011).
Update on the Safety of Phosphodiesterase Type 5 Inhibitors for the Treatment of Erectile Dysfunction
2018, Sexual Medicine ReviewsCitation Excerpt :The overwhelming majority of pills was obtained without prescription and was used concomitantly with alcohol and illicit drugs, particularly marijuana and amphetamines.90 Case reports of seizures and myocardial infarction have been reported in these men, likely potentiated by the vasodilatory, pro-convulsant, and cytochrome P450 inhibitory properties of these illicit drugs.91–94 This recreational use also represents a public health concern because it has been associated with a higher incidence of sexually transmitted infections, including HIV, particularly when associated with illicit drug use.95,96
